Envelopement and secretion of CCHFV
Crimean-Congo hemorrhagic fever virus (CCHFV) is an emerging tick-borne pathogen that causes hemorrhagic disease in humans (mortality rate up to 30%). In 2023, this virus was detected for the first time in France in ticks isolated from cattle in southern France and Corsica. Due to the confinement imposed by the high pathogenicity of this virus (BSL4), few molecular data exist on its assembly mechanisms.
Using a system based on the production of transcriptionally competent viral particles (tcVLPs), we have identified and characterized various motifs that govern the trafficking of Gn and Gc envelope glycoproteins to CCHFV assembly sites. Using the full length virus, we also identified the PACS-2 protein (Phosphofurin acidic cluster sorting protein) as an cellular host factor for the envelopment and secretion of viral particles.
