Abstract: We have established an in situ cryo-correlative light and electron tomography workflow to study cellular organelles are their interactions with viruses during infection in unperturbed conditions and at a molecular resolution. This method allowed us to describe the architecture of lamellar bodies present in type II pneumocytes and to capture influenza A virus entry intermediates. In addition, our data revealed a viral RNA network inside double-membrane vesicles and assembly steps of SARS-CoV-2.
