Abstract: Viral emergence is a complex process in which viruses must overcome numerous ecological, immunological, and molecular barriers. While metagenomics has greatly expanded our understanding of viral diversity in animal reservoirs, the potential threat posed by these newly identified viruses to human health remains largely unknown-partly due to limited functional characterization. Here, we employed large-scale viral pseudotyping to assess the ability of a broad panel of enveloped RNA viruses to enter human cells. We then identified viral features and host molecular determinants that govern the human cellular tropism of animal viruses, allowing the identification of new viral entry factors.
