Abstract: Rhabdoviruses first enter the cell via the endocytic pathway and then fuse with the cell membrane in the acidic environment of the endosome. Receptor recognition and membrane fusion are mediated by a single viral transmembrane glycoprotein (G). The fusion process is triggered by a structural rearrangement of G induced by low pH. G is an atypical fusion protein in that there is a pH-dependent equilibrium between its pre- and post-fusion conformations. We will first give an overview on Vesiculovirus G structural data, which is one of the best characterized viral fusion glycoproteins to date. This will be followed by an insight into glycoproteins of the Lyssavirus genus (related to the rabies virus). Understanding the entry pathway of rhabdoviruses could have a major impact on biotechnologies as, for example, Vesiculovirus G is widely used to pseudotype lentiviruses to promote efficient transduction of target cells.
